(Pure Culture)

(Fermenter)

Chapter 14 — Science & Technology

Fermentation Technology

RAS Mains — General Science & Technology |

Objective: Fermentation technology should not be understood merely as "the process of making alcohol from sugar." In modern biotechnology, fermentation is used to produce food products, ethanol, organic acids, antibiotics, enzymes, vitamins, biofuels, and other industrial products. Therefore, in this chapter, it is essential to understand the complete sequence from biological principles to industrial fermenters and product separation.

1 What is Fermentation?

Fermentation is a biochemical process in which microorganisms or their cells, with the help of enzymes, transform organic substances, especially sugars, under controlled conditions to produce useful products.

In the traditional sense, fermentation is often referred to as a process of obtaining energy in the absence of oxygen, but in industrial biotechnology, its meaning is broader.

Simple Flow

Raw material → Microorganism/Cell → Controlled fermentation → Product → Separation and purification

Example: Glucose → Yeast → Ethanol + CO₂

2 Basic Scientific Basis of Fermentation

Microorganisms require energy and nutrients for growth and metabolism.

When suitable organic matter is provided, microorganisms use it to:

Therefore, in industrial fermentation, not only the microorganism is important, but the microorganism + nutrient medium + temperature + pH + oxygen + time + fermenter are all important.

3 Major Microorganisms Used in Fermentation

3.1 Yeast by RAS A2Z

Major example— Saccharomyces cerevisiae

Main uses—

3.2 Bacteria by RAS A2Z

Different bacteria are used for different products.

Examples—

3.3 Fungi by RAS A2Z

Fungi are extremely important in industrial fermentation.

Examples—

4 Major Types of Fermentation

Fermentation can be classified on various bases.

4.1 Based on Oxygen by RAS A2Z

(1) Anaerobic Fermentation

The process occurs in the absence or very low availability of oxygen.

Example: Ethanol production by yeast

C₆H₁₂O₆ → 2C₂H₅OH + 2CO₂ + Energy
Glucose → Ethanol + Carbon dioxide

(2) Aerobic/Oxygen-requiring Fermentation

In some industrial processes, oxygen is required for the growth of microorganisms or product formation.

Example: Ethanol → Acetic acid

This process occurs by Acetobacter in the presence of oxygen.

5 Alcoholic Fermentation

This is one of the most famous fermentation processes.

Microorganism— Saccharomyces cerevisiae

Raw materials—

Main products— Ethanol, Carbon dioxide

C₆H₁₂O₆ → 2C₂H₅OH + 2CO₂

Uses—

6 Lactic Acid Fermentation

In this, mainly lactic acid is formed from sugar.

Major microorganisms— Lactobacillus and other lactic acid bacteria.

Main uses—

When lactose in milk is converted to lactic acid, the pH decreases and the milk protein begins to coagulate.

7 Acetic Acid Fermentation

Acetic acid is the main acidic component of vinegar.

Major microorganism— Acetobacter

It oxidizes ethanol to form acetic acid.

Ethanol + O₂ → Acetic acid + Water

Uses—

📌 Exam Fact

Alcoholic fermentation → Ethanol

Acetic fermentation → Acetic acid

8 Fermentation in Bread Making

Yeast is used in bread making.

Process

Dough + Water + Yeast + Suitable conditions

↓

Yeast ferments sugar

↓

CO₂ is produced

↓

Gas bubbles form in the dough

↓

The dough rises

↓

After baking, bread is ready.

📌 Important Fact

The main reason for bread rising is CO₂ gas, not ethanol.

9 Fermentation Medium

The liquid or medium that provides nutrition to microorganisms in a fermenter is called the fermentation medium.

Depending on the requirement, it may contain—

Carbon Sources

Nitrogen Sources

10 Substrate

The substance on which microorganisms act to produce a product is called the substrate.

Example: Glucose → Yeast → Ethanol

Here, glucose is the substrate.

In industrial production, a cheap and easily available substrate is economically important.

11 Inoculum

The active microbial culture added to the fermenter to start fermentation is called the inoculum.

A good inoculum should have—

12 Pure Culture

In industrial fermentation, the importance of a pure culture of the desired microorganism is extremely high.

If other unwanted microorganisms enter, they can:

Therefore, sterility and contamination control are important in industrial fermentation.

13 Fermenter

The equipment in which industrial fermentation is carried out under controlled conditions is called a fermenter or bioreactor.

It is not just a tank, but a controlled biological reaction system.

Major Components

  1. Vessel/Tank
  2. Stirrer
  3. Air sparger
  4. Temperature control
  5. pH control
  6. Sensors
  7. Foam control
  8. Nutrient inlet system
  9. Sampling system
  10. Exhaust system

14 Stirrer of the Fermenter

The function of the stirrer is to mix the fermentation medium.

This ensures—

15 Air Sparger

Oxygen may be required in aerobic fermentation.

The air sparger functions to introduce bubbles of air/oxygen into the medium. This provides the necessary oxygen to the microorganisms.

16 Importance of Temperature

Each microorganism has a suitable temperature range.

Too high temperature:

Too low temperature:

Therefore, temperature control is essential in industrial fermentation.

17 Importance of pH

The growth of microorganisms and enzyme activity are affected by pH.

If the pH goes outside the suitable range, then:

Therefore, pH can be controlled in the fermenter by sensors and controllers.

18 Importance of Oxygen

In fermentation, the requirement for oxygen depends on the product and the microorganism.

Examples—

Therefore, saying "fermentation always occurs without oxygen" is an incomplete statement in the industrial context.

19 Foam Control

In some fermentation processes, foam can form due to proteins, cellular substances, and air.

Excessive foam:

Therefore, antifoam agents or mechanical foam control can be used.

20 Major Steps of the Fermentation Process

Industrial fermentation can generally be understood in the following sequence:

  1. Step 1 — Selection of microorganism
  2. Step 2 — Preservation of pure culture

  7. Step 7 — Controlled fermentation
  8. Step 8 — Product formation
  9. Step 9 — Product separation
  10. Step 10 — Purification
  11. Step 11 — Preparation of final product

21 Sterilization

Before fermentation, it is important to free the medium and equipment from unwanted microorganisms.

Objectives—

Methods—
