उद्देश्य: किणवन तकनीकी को केवल "शर्करा से अल्कोहल बनने की प्रक्रिया" तक सीमित नहीं समझना चाहिए। आधुनिक जैव-प्रौद्योगिकी (Biotechnology) में किणवन द्वारा खाद्य पदार्थ, एथेनॉल, कार्बनिक अम्ल, एंटीबायोटिक, एंजाइम, विटामिन, जैव-ईंधन तथा अन्य औद्योगिक उत्पाद बनाए जाते हैं। इसलिए इस अध्याय में जैविक सिद्धांत से लेकर औद्योगिक फर्मेंटर और उत्पाद पृथक्करण तक पूरा क्रम समझना आवश्यक है।
1 किणवन क्या है?
किणवन (Fermentation) एक जैव-रासायनिक प्रक्रिया है जिसमें सूक्ष्मजीव (Microorganisms) या उनकी कोशिकाएँ एंजाइमों (Enzymes) की सहायता से कार्बनिक पदार्थों, विशेषकर शर्करा, का नियंत्रित परिस्थितियों में रूपांतरण करके उपयोगी उत्पाद बनाती हैं।
पारंपरिक अर्थ में किणवन प्रायः ऑक्सीजन की अनुपस्थिति में ऊर्जा प्राप्त करने वाली प्रक्रिया को कहा जाता है, लेकिन औद्योगिक जैव-प्रौद्योगिकी में इसका अर्थ अधिक व्यापक है।

उदाहरण: ग्लूकोज → यीस्ट → एथेनॉल + CO₂
2 किणवन का मूल वैज्ञानिक आधार
सूक्ष्मजीवों को वृद्धि और उपापचय (Metabolism) के लिए ऊर्जा एवं पोषक तत्वों की आवश्यकता होती है।
जब उपयुक्त कार्बनिक पदार्थ उपलब्ध कराया जाता है, तो सूक्ष्मजीव उसका उपयोग करके:
- ऊर्जा प्राप्त करते हैं।
- अपनी कोशिकीय वृद्धि करते हैं।
- नए जैव-अणु बनाते हैं।
- विशिष्ट औद्योगिक उत्पाद उत्पन्न करते हैं।
इसलिए औद्योगिक किणवन में केवल सूक्ष्मजीव ही महत्वपूर्ण नहीं है, बल्कि सूक्ष्मजीव + पोषक माध्यम + तापमान + pH + ऑक्सीजन + समय + फर्मेंटर सभी महत्वपूर्ण हैं।
3 किणवन में प्रयुक्त प्रमुख सूक्ष्मजीव
3.1 यीस्ट (Yeast) by RAS A2Z
प्रमुख उदाहरण— Saccharomyces cerevisiae
मुख्य उपयोग—
- एथेनॉल उत्पादन
- ब्रेड निर्माण
- जैव-ईंधन
- कुछ औद्योगिक जैव-उत्पाद
3.2 जीवाणु (Bacteria) by RAS A2Z
विभिन्न जीवाणुओं का उपयोग अलग-अलग उत्पादों के लिए किया जाता है।
उदाहरण—
- Lactobacillus → लैक्टिक अम्ल
- Acetobacter → एसीटिक अम्ल/सिरका
- Bacillus → विभिन्न एंजाइम एवं जैव-उत्पाद
- अन्य औद्योगिक जीवाणु → कार्बनिक अम्ल, एंजाइम आदि
3.3 कवक (Fungi) by RAS A2Z
कवक औद्योगिक किणवन में अत्यंत महत्वपूर्ण हैं।
उदाहरण—
- Aspergillus niger → साइट्रिक अम्ल
- Penicillium → पेनिसिलिन
- अन्य कवक → एंजाइम, कार्बनिक अम्ल एवं जैव-सक्रिय पदार्थ
4 किणवन के प्रमुख प्रकार
किणवन को विभिन्न आधारों पर वर्गीकृत किया जा सकता है।
4.1 ऑक्सीजन के आधार पर by RAS A2Z
(1) अवायवीय किणवन (Anaerobic Fermentation)
ऑक्सीजन की अनुपस्थिति या बहुत कम उपलब्धता में प्रक्रिया होती है।
उदाहरण: यीस्ट द्वारा एथेनॉल निर्माण
ग्लूकोज → एथेनॉल + कार्बन डाइऑक्साइड
(2) वायवीय/ऑक्सीजनी परिस्थितियों वाला किणवन
कुछ औद्योगिक प्रक्रियाओं में सूक्ष्मजीवों की वृद्धि या उत्पाद निर्माण के लिए ऑक्सीजन की आवश्यकता होती है।
उदाहरण: एथेनॉल → एसीटिक अम्ल
यह प्रक्रिया Acetobacter द्वारा ऑक्सीजन की उपस्थिति में होती है।
5 अल्कोहलिक किणवन (Alcoholic Fermentation)
यह सबसे प्रसिद्ध किणवन प्रक्रियाओं में से एक है।
सूक्ष्मजीव— Saccharomyces cerevisiae
कच्चा पदार्थ—
- ग्लूकोज
- गन्ने का रस
- शीरा (Molasses)
- स्टार्च से प्राप्त शर्करा
मुख्य उत्पाद— एथेनॉल, कार्बन डाइऑक्साइड
उपयोग—
- औद्योगिक अल्कोहल
- जैव-ईंधन
- एथेनॉल मिश्रित पेट्रोल
- ब्रेड उद्योग
6 लैक्टिक अम्ल किणवन
इसमें शर्करा से मुख्यतः लैक्टिक अम्ल (Lactic acid) बनता है।
प्रमुख सूक्ष्मजीव— Lactobacillus तथा अन्य लैक्टिक अम्ल जीवाणु।
प्रमुख उपयोग—
- दही
- किण्वित खाद्य पदार्थ
- लैक्टिक अम्ल उत्पादन
- खाद्य उद्योग
दूध में लैक्टोज का लैक्टिक अम्ल में परिवर्तन होने से pH घटता है और दूध का प्रोटीन जमने लगता है।
7 एसीटिक अम्ल किणवन
एसीटिक अम्ल (Acetic acid) सिरके का प्रमुख अम्लीय घटक है।
प्रमुख सूक्ष्मजीव— Acetobacter
यह एथेनॉल को ऑक्सीकरण करके एसीटिक अम्ल बनाता है।
उपयोग—
- सिरका निर्माण
- खाद्य परिरक्षण
- खाद्य उद्योग

8 ब्रेड निर्माण में किणवन
ब्रेड निर्माण में यीस्ट का उपयोग किया जाता है।
प्रक्रिया
आटा + जल + यीस्ट + उपयुक्त परिस्थितियाँ
↓
यीस्ट शर्करा का किण्वन करता है
↓
CO₂ उत्पन्न होती है
↓
आटे में गैस के बुलबुले बनते हैं

आटा फूलता है
↓
बेकिंग के बाद ब्रेड तैयार होती है।
ब्रेड के फूलने का मुख्य कारण CO₂ गैस है, न कि एथेनॉल।
9 किणवन माध्यम (Fermentation Medium)
फर्मेंटर में सूक्ष्मजीवों को पोषण देने वाले द्रव या माध्यम को किणवन माध्यम कहा जाता है।
इसमें आवश्यकता के अनुसार—
- कार्बन स्रोत
- नाइट्रोजन स्रोत
- खनिज
- लवण
- विटामिन
- जल
- अन्य वृद्धि कारक
हो सकते हैं।
कार्बन स्रोत
- ग्लूकोज
- सुक्रोज
- शीरा

10 सब्सट्रेट (Substrate)
जिस पदार्थ पर सूक्ष्मजीव क्रिया करके उत्पाद बनाते हैं, उसे सब्सट्रेट (Substrate) कहा जाता है।
उदाहरण: ग्लूकोज → यीस्ट → एथेनॉल
यहाँ ग्लूकोज सब्सट्रेट है।
औद्योगिक उत्पादन में सस्ता और आसानी से उपलब्ध सब्सट्रेट आर्थिक दृष्टि से महत्वपूर्ण होता है।
11 इनोकुलम (Inoculum)
किणवन शुरू करने के लिए फर्मेंटर में डाली जाने वाली सक्रिय सूक्ष्मजीवी संस्कृति को इनोकुलम (Inoculum) कहते हैं।
अच्छे इनोकुलम में—
- वांछित सूक्ष्मजीव
- पर्याप्त संख्या
- सक्रिय अवस्था
- कम संदूषण (Contamination)
- उचित वृद्धि क्षमता
होनी चाहिए।
12 शुद्ध संवर्धन (Pure Culture)
औद्योगिक किणवन में इच्छित सूक्ष्मजीव की शुद्ध संस्कृति (Pure culture) का महत्व अत्यधिक है।
यदि अन्य अवांछित सूक्ष्मजीव प्रवेश कर जाएँ तो:
- पोषक पदार्थों की प्रतिस्पर्धा बढ़ सकती है।
- उत्पाद की मात्रा घट सकती है।
- अवांछित उत्पाद बन सकते हैं।
- पूरी प्रक्रिया प्रभावित हो सकती है।
इसलिए औद्योगिक किणवन में स्टेरिलिटी (Sterility) और संदूषण नियंत्रण महत्वपूर्ण हैं।
13 फर्मेंटर (Fermenter)
जिस उपकरण में नियंत्रित परिस्थितियों में औद्योगिक किणवन कराया जाता है, उसे फर्मेंटर (Fermenter) या बायोरिएक्टर (Bioreactor) कहते हैं।
यह केवल एक टैंक नहीं है, बल्कि एक नियंत्रित जैव-प्रतिक्रिया प्रणाली है।

14 फर्मेंटर का स्टिरर
Stirrer का कार्य किणवन माध्यम को मिलाना है।
इससे—
- पोषक तत्व समान रूप से वितरित होते हैं।
- तापमान समान रहता है।
- सूक्ष्मजीव पूरे माध्यम में फैलते हैं।
- ऑक्सीजन की आवश्यकता होने पर उसका स्थानांतरण बेहतर होता है।
15 एयर स्पार्जर (Air Sparger)
एरोबिक किणवन में ऑक्सीजन आवश्यक हो सकती है।
Air sparger माध्यम में वायु/ऑक्सीजन के बुलबुले प्रविष्ट कराने का कार्य करता है। इससे सूक्ष्मजीवों को आवश्यक ऑक्सीजन उपलब्ध होती है।
16 तापमान का महत्व
प्रत्येक सूक्ष्मजीव की एक उपयुक्त तापमान सीमा होती है।
बहुत अधिक तापमान:
- एंजाइमों को निष्क्रिय कर सकता है।
- कोशिकाओं को क्षति पहुँचा सकता है।
बहुत कम तापमान:
- सूक्ष्मजीवी वृद्धि धीमी कर सकता है।
- उत्पाद निर्माण की दर घटा सकता है।
इसलिए औद्योगिक किणवन में तापमान का नियंत्रण आवश्यक है।
17 pH का महत्व
सूक्ष्मजीवों की वृद्धि और एंजाइम गतिविधि pH से प्रभावित होती है।
यदि pH उपयुक्त सीमा से बाहर चला जाए तो:
- वृद्धि कम हो सकती है।
- उत्पाद निर्माण घट सकता है।
- अवांछित उपापचयी उत्पाद बन सकते हैं।
इसलिए फर्मेंटर में pH को सेंसर और नियंत्रक द्वारा नियंत्रित किया जा सकता है।
18 ऑक्सीजन का महत्व
किणवन में ऑक्सीजन की आवश्यकता उत्पाद और सूक्ष्मजीव पर निर्भर करती है।
उदाहरण—
- यीस्ट का अल्कोहलिक किणवन → कम/अवायवीय परिस्थितियों में एथेनॉल उत्पादन।
- Acetobacter द्वारा एसीटिक अम्ल उत्पादन → ऑक्सीजन आवश्यक।
इसलिए "किणवन हमेशा बिना ऑक्सीजन होता है" कहना औद्योगिक संदर्भ में अधूरा कथन है।
19 फोम (Foam) नियंत्रण
कुछ किणवन प्रक्रियाओं में प्रोटीन, कोशिकीय पदार्थ और वायु के कारण फोम बन सकता है।
अत्यधिक फोम:
- उपकरण से बाहर निकल सकता है।
- संदूषण का जोखिम बढ़ा सकता है।
- प्रक्रिया की निगरानी में बाधा डाल सकता है।
इसलिए antifoam agents या यांत्रिक फोम नियंत्रण का प्रयोग किया जा सकता है।
20 किणवन प्रक्रिया के प्रमुख चरण
औद्योगिक किणवन को सामान्यतः निम्न क्रम में समझा जा सकता है:
- चरण 1 — सूक्ष्मजीव का चयन
- चरण 2 — शुद्ध संस्कृति का संरक्षण

21 स्टेरिलीकरण (Sterilization)
किणवन से पहले माध्यम और उपकरणों को अवांछित सूक्ष्मजीवों से मुक्त करना महत्वपूर्ण है।
उद्देश्य—
- संदूषण रोकना
- उत्पाद की शुद्धता बनाए रखना
- वांछित सूक्ष्मजीव को प्रतिस्पर्धा से बचाना
विधियाँ—
- ऊष्मीय स्टेरिलीकरण
- भाप द्वारा स्टेरिलीकरण
- फिल्ट्रेशन, जहाँ उपयुक्त हो
- उपकरणों की स्वच्छता
22 बैच किण्वन (Batch Fermentation)
इसमें शुरुआत में आवश्यक माध्यम फर्मेंटर में डाल दिया जाता है और प्रक्रिया पूरी होने तक उत्पादन किया जाता है।
विशेषताएँ—
- सरल संचालन
- अपेक्षाकृत आसान नियंत्रण
- प्रक्रिया समाप्त होने पर उत्पाद निकाला जाता है।
उपयोग: कई औद्योगिक जैव-उत्पादों में बैच प्रक्रिया उपयोगी होती है।
23 फेड-बैच किण्वन (Fed-Batch Fermentation)
इसमें प्रक्रिया के दौरान पोषक पदार्थों को नियंत्रित मात्रा में धीरे-धीरे जोड़ा जाता है।
लाभ—
- पोषक तत्वों का बेहतर नियंत्रण
- अत्यधिक सब्सट्रेट सांद्रता से बचाव
- उत्पादकता में सुधार की संभावना
यह औद्योगिक जैव-प्रक्रियाओं में महत्वपूर्ण तकनीक है।
24 सतत किण्वन (Continuous Fermentation)
इसमें ताजा माध्यम लगातार या नियंत्रित दर से फर्मेंटर में प्रवेश करता है और उत्पाद युक्त माध्यम लगातार बाहर निकलता रहता है।
विशेषताएँ—
- निरंतर संचालन
- उच्च उत्पादकता की संभावना
- प्रक्रिया नियंत्रण अधिक जटिल

25 प्राथमिक एवं द्वितीयक उपापचयी उत्पाद
सूक्ष्मजीवों द्वारा बनाए गए उत्पादों को उपापचय के आधार पर समझना उपयोगी है।
प्राथमिक उपापचयी उत्पाद (Primary metabolites)
ये सामान्यतः सक्रिय वृद्धि के दौरान बनते हैं।
उदाहरण— कुछ अमीनो अम्ल, कार्बनिक अम्ल, एथेनॉल।
द्वितीयक उपापचयी उत्पाद (Secondary metabolites)
ये सामान्य वृद्धि के लिए हमेशा आवश्यक नहीं होते और अक्सर वृद्धि के बाद के चरणों में बनते हैं।
उदाहरण— कुछ एंटीबायोटिक, जैव-सक्रिय यौगिक।
सभी किणवन उत्पाद केवल एथेनॉल नहीं होते।
26 उत्पाद पृथक्करण — डाउनस्ट्रीम प्रोसेसिंग
किणवन के बाद प्राप्त मिश्रण से उपयोगी उत्पाद को अलग एवं शुद्ध करने की प्रक्रिया को डाउनस्ट्रीम प्रोसेसिंग (Downstream processing) कहते हैं।
इसमें आवश्यकता के अनुसार—
- कोशिका पृथक्करण
- फिल्ट्रेशन
- सेंट्रीफ्यूगेशन
- निष्कर्षण

27 अपस्ट्रीम प्रोसेसिंग (Upstream Processing)
किणवन शुरू होने से पहले की तैयारियों को व्यापक रूप से अपस्ट्रीम प्रोसेसिंग (Upstream processing) कहा जाता है।
इसमें—
- सूक्ष्मजीव चयन
- संस्कृति संरक्षण
- माध्यम निर्माण
- इनोकुलम विकास
- स्टेरिलीकरण
- प्रक्रिया परिस्थितियों की तैयारी
शामिल हैं।
