1 जैव-प्रौद्योगिकी क्या है?
जैव-प्रौद्योगिकी (Biotechnology) वह तकनीकी क्षेत्र है जिसमें जीवित जीवों, कोशिकाओं, एंजाइमों या जैविक प्रक्रियाओं का उपयोग करके मानव उपयोग के लिए उपयोगी उत्पाद एवं प्रक्रियाएँ विकसित की जाती हैं।

3 आनुवंशिक इंजीनियरिंग क्या है?
आनुवंशिक इंजीनियरिंग (Genetic Engineering) वह तकनीक है जिसमें किसी जीव के DNA या जीन में लक्षित परिवर्तन करके वांछित गुण प्राप्त करने का प्रयास किया जाता है।
इसमें—
जैसी प्रक्रियाएँ शामिल हो सकती हैं।
4 DNA — आनुवंशिक पदार्थ
DNA (Deoxyribonucleic Acid) अधिकांश जीवों में आनुवंशिक सूचना का प्रमुख वाहक है।

प्रत्येक न्यूक्लियोटाइड में—
- नाइट्रोजनयुक्त क्षारक
- डीऑक्सीराइबोज शर्करा

5 जीन क्या है?
जीन (Gene) DNA का वह विशिष्ट अनुक्रम है जिसमें किसी कार्यात्मक RNA या प्रोटीन से संबंधित आनुवंशिक सूचना निहित होती है।
हालाँकि सभी जीन सीधे प्रोटीन नहीं बनाते; कुछ कार्यात्मक RNA भी बनाते हैं।
6 जीन अभिव्यक्ति (Gene Expression)
किसी जीन में निहित सूचना का कार्यात्मक उत्पाद में उपयोग होना जीन अभिव्यक्ति कहलाता है।

7 Recombinant DNA क्या है?
जब अलग-अलग स्रोतों से प्राप्त DNA अनुक्रमों को जोड़कर एक नई DNA संरचना बनाई जाती है, तो उसे पुनर्संयोजित DNA (Recombinant DNA) कहते हैं।

8 Recombinant DNA Technology
पुनर्संयोजित DNA तकनीक में सामान्यतः निम्न चरण समझे जाते हैं:
- चरण 1 — वांछित जीन की पहचान।
- चरण 2 — DNA का पृथक्करण।
- चरण 3 — वांछित DNA अनुक्रम को काटना।
- चरण 4 — वेक्टर (Vector) में जीन जोड़ना।
- चरण 5 — Recombinant DNA बनाना।
- चरण 6 — इसे होस्ट कोशिका में प्रवेश कराना।
- चरण 7 — जीन की अभिव्यक्ति।
- चरण 8 — उत्पाद प्राप्त करना और शुद्ध करना।
9 Restriction Enzymes
प्रतिबंधक एंजाइम (Restriction Enzymes) DNA को विशिष्ट न्यूक्लियोटाइड अनुक्रमों पर काटने वाले एंजाइम हैं।
इन्हें आनुवंशिक इंजीनियरिंग की "molecular scissors" कहा जाता है।
उदाहरण: EcoRI
यह DNA को एक विशिष्ट पहचान अनुक्रम पर काटता है। कुछ restriction enzymes ऐसे कट बनाते हैं जिनसे sticky ends बनते हैं।
10 DNA Ligase
DNA Ligase DNA के टूटे हुए भागों को जोड़ने वाला एंजाइम है।
आनुवंशिक इंजीनियरिंग में—
- Restriction enzyme → DNA काटता है
- DNA ligase → DNA जोड़ता है
इसी कारण दोनों recombinant DNA निर्माण में अत्यंत महत्वपूर्ण हैं।
11 वेक्टर (Vector)
वह DNA अणु जो वांछित जीन को होस्ट कोशिका तक पहुँचाने में सहायता करता है, वेक्टर कहलाता है।

12 प्लास्मिड (Plasmid)
प्लास्मिड छोटे, सामान्यतः वृत्ताकार, अतिरिक्त-गुणसूत्रीय DNA अणु होते हैं जो विशेषकर बैक्टीरिया में पाए जा सकते हैं।
आनुवंशिक इंजीनियरिंग में प्लास्मिड को वेक्टर के रूप में उपयोग किया जाता है। इसमें वांछित जीन जोड़कर recombinant DNA बनाया जा सकता है।
13 Host Cell
जिस कोशिका में recombinant DNA पहुँचाया जाता है और जहाँ उसका प्रतिकृतिकरण या अभिव्यक्ति होती है, उसे Host Cell कहते हैं।

14 Transformation
जब बाहरी DNA को किसी कोशिका, विशेषकर बैक्टीरिया, में प्रवेश कराया जाता है और कोशिका उसे ग्रहण करती है, तो इस प्रक्रिया को Transformation कहा जाता है। यह recombinant DNA technology का महत्वपूर्ण चरण है।
15 चयन योग्य मार्कर (Selectable Marker)
Recombinant DNA प्राप्त करने वाली कोशिकाओं की पहचान करने के लिए Selectable marker का उपयोग किया जा सकता है।
इससे वैज्ञानिक यह पहचान सकते हैं कि कौन-सी कोशिकाएँ वांछित DNA लेकर गई हैं।

16 PCR क्या है?
PCR (Polymerase Chain Reaction) किसी विशिष्ट DNA अनुक्रम की बहुत बड़ी संख्या में प्रतियाँ बनाने की तकनीक है।
मुख्य चरण
- Denaturation: DNA की दोहरी कुंडली अलग होती है।
- Annealing: Primers लक्षित DNA अनुक्रम से जुड़ते हैं।
- Extension: DNA polymerase नया DNA बनाता है।
यह चक्र कई बार दोहराया जाता है।
17 PCR में Taq Polymerase
PCR में सामान्य DNA polymerase की बजाय ऊष्मा-सहिष्णु Taq DNA polymerase का उपयोग किया जाता है।
यह मूलतः Thermus aquaticus नामक ऊष्माप्रिय जीवाणु से संबंधित है।

18 DNA Sequencing
DNA में न्यूक्लियोटाइडों के क्रम को निर्धारित करने की प्रक्रिया को DNA sequencing कहते हैं।
उदाहरण: A–T–G–C–C–A… का सही क्रम निर्धारित करना।

19 जीन क्लोनिंग (Gene Cloning)
किसी विशिष्ट DNA अनुक्रम की अनेक समान प्रतियाँ बनाना जीन क्लोनिंग कहलाता है।
इससे किसी विशिष्ट जीन का अध्ययन या उसका उत्पाद प्राप्त करना संभव होता है।
20 जीन संपादन (Genome Editing)
Genome Editing में DNA के किसी विशिष्ट स्थान पर लक्षित परिवर्तन किया जाता है।
इसमें—
- DNA का कुछ भाग हटाना
- नया अनुक्रम जोड़ना
- किसी आधार में परिवर्तन
- जीन की कार्यक्षमता बदलना
जैसे परिवर्तन संभव हो सकते हैं।
21 CRISPR-Cas9
CRISPR-Cas9 आधुनिक genome-editing की प्रमुख तकनीकों में से एक है।

25 वैक्सीन उत्पादन
आधुनिक जैव-प्रौद्योगिकी द्वारा विभिन्न प्रकार की वैक्सीन विकसित की जा सकती हैं।
तकनीकों में—

26 जीन थेरेपी
जीन थेरेपी (Gene Therapy) में रोग के उपचार के लिए कोशिकाओं के आनुवंशिक पदार्थ में चिकित्सकीय उद्देश्य से परिवर्तन किया जाता है।
उद्देश्य हो सकते हैं—
- दोषपूर्ण जीन के प्रभाव को कम करना
- कार्यात्मक आनुवंशिक सामग्री उपलब्ध कराना
- रोग पैदा करने वाली प्रक्रिया को लक्षित करना
Somatic Gene Therapy
परिवर्तन शरीर की गैर-जनन कोशिकाओं में किया जाता है और सामान्यतः अगली पीढ़ी को विरासत में नहीं जाता।
27 Germline Editing
जनन कोशिकाओं या ऐसे प्रारंभिक भ्रूणीय चरणों में आनुवंशिक परिवर्तन जिनसे परिवर्तन अगली पीढ़ियों तक पहुँच सकता है, germline/heritable editing से संबंधित है।
यह क्षेत्र वैज्ञानिक के साथ-साथ:

- सामाजिक
- कानूनी
- सुरक्षा
प्रश्न भी उत्पन्न करता है।
WHO ने human genome editing के लिए विशेष governance और oversight की आवश्यकता पर बल दिया है तथा heritable human genome editing को गंभीर नैतिक एवं सुरक्षा चिंताओं वाला क्षेत्र माना है।

कृषि जैव-प्रौद्योगिकी के प्रमुख उद्देश्य:

29 Bt फसल
Bt crops में Bacillus thuringiensis से संबंधित जीन का उपयोग कीट-प्रतिरोधी गुण विकसित करने के लिए किया गया है।
Bt toxin
कुछ Bt प्रोटीन कीटों के विशिष्ट लार्वा पर विषाक्त प्रभाव डाल सकते हैं।

इसका उद्देश्य कपास के कुछ प्रमुख कीटों से सुरक्षा प्रदान करना है।
30 गोल्डन राइस और जैव-सुदृढ़ीकरण
Golden Rice को विटामिन A के पूर्वगामी β-carotene के उत्पादन से संबंधित आनुवंशिक संशोधन के उदाहरण के रूप में पढ़ाया जाता है।

विशेष रूप से आनुवंशिक रूप से संशोधित जीवों तथा GM crops के मामले में जोखिम मूल्यांकन और नियामक प्रक्रियाएँ महत्वपूर्ण हैं। ICAR ने GM crops के संदर्भ में biosafety, regulatory aspects तथा field trials जैसे विषयों को प्रमुख माना है।
42 CRISPR और भारत — नवीन अनुप्रयोग
भारत में genome editing का कृषि क्षेत्र में वास्तविक विकास देखने को मिला है। ICAR ने 2025 में DRR Rice 100 (Kamla) और Pusa DST Rice 1 नामक genome-edited rice varieties की घोषणा की।
ICAR के अनुसार ये CRISPR-Cas आधारित genome-editing से विकसित की गई और इनमें उपज, जल उपयोग तथा जलवायु-सहनशीलता से संबंधित लाभ बताए गए हैं।
यह तथ्य RAS के लिए समसामयिक + विज्ञान + कृषि के intersection पर अत्यंत उपयोगी है।

Biotechnology is a technological field in which living organisms, cells, enzymes, or biological processes are used to develop useful products and processes for human use.

8 Recombinant DNA Technology
In recombinant DNA technology, the following steps are generally understood:
- Step 1 — Identification of desired gene.

- Step 5 — Making recombinant DNA.
- Step 6 — Introducing it into the host cell.

9 Restriction Enzymes
Restriction enzymes are enzymes that cut DNA at specific nucleotide sequences.
They are called "molecular scissors" of genetic engineering.
Example: EcoRI
It cuts DNA at a specific recognition sequence. Some restriction enzymes make cuts that produce sticky ends.
10 DNA Ligase
DNA Ligase is an enzyme that joins broken parts of DNA.
In genetic engineering—

- DNA ligase → joins DNA
Therefore, both are extremely important in the construction of recombinant DNA.
11 Vector
The DNA molecule that helps carry the desired gene to the host cell is called a vector.

12 Plasmid
Plasmids are small, generally circular, extra-chromosomal DNA molecules found especially in bacteria.
In genetic engineering, plasmids are used as vectors. By inserting the desired gene into them, recombinant DNA can be made.
13 Host Cell
The cell into which recombinant DNA is introduced and where it replicates or expresses is called the Host Cell.

14 Transformation
When external DNA is introduced into a cell, especially bacteria, and the cell takes it up, this process is called Transformation. This is an important step of recombinant DNA technology.
15 Selectable Marker
Selectable markers can be used to identify cells that have received recombinant DNA.
This helps scientists identify which cells have taken up the desired DNA.

16 What is PCR?
PCR (Polymerase Chain Reaction) is a technique for making a very large number of copies of a specific DNA sequence.

- Denaturation: The double helix of DNA separates.

- Extension: DNA polymerase synthesizes new DNA.
This cycle is repeated many times.
17 Taq Polymerase in PCR
In PCR, instead of ordinary DNA polymerase, heat-tolerant Taq DNA polymerase is used.
It is originally related to the thermophilic bacterium Thermus aquaticus.

The process of determining the order of nucleotides in DNA is called DNA sequencing.
